Yes, (many people use *BSD boxes for ethernet to ethenet routers) you'll
probably need to build a custom kernel with modified device entries for
the multiple cards.

If the cards are software configurable you'll have to configure each
one separatly, with only one in the machine at a time.  Then just
create the relevent device entries in the kernel config file.

If there is one entry for your card with say:

  device ep0 at isa? port 0x300 net irq 10 vector epintr

and you have another card of the same type then just add another entry, eg:

  device ep1 at isa? port 0x330 net irq 11 vector epintr
